- Bioactive properties of the endemic algerian Myrtus nivelii Batt & Trab.: scientific approaches to the traditional usesPublication . In this context, bioactivities of the aqueous extract and its organic sub-fractions (ethyl acetate and butanol) were evaluated with focus onantioxidant properties (free radicals’ scavenging activity, reducing power, inhibition of β-carotene blanching andlipid peroxidation), anti-inflammatory potential. (inhibition of NO production in lipopolysaccharide-stimulated RAW 264.7 macrophages), cytotoxicity for human tumor cells and normal porcine liver primary cells, and antibacterial activity against ten clinical isolates. The obtained results indicated high activities for all the tested samples. The antioxidant activity was higher than that obtained for Trolox (positive control), showing the ethylacetate fraction the highest activity as also the highest inhibition of NO production (EC50= 104 ± 6 μg/mL) and cytotoxicity for all the tested tumor cell lines (HeLa- 15.39±1 μg/mL, MCF-7- 16.12± 1μg/mL, NCI-H460-, HepG2-17.67±1 μg/mL and 49.01 ± 3 μg/mL). The butanol fraction and the crude aqueous extract presented the highest antibacterial activity. Overall, this study highlights the potential of M. nivellii leaves in the preparation of bioactive ingredients.

Rached, Wahiba; Bennaceur, Malika; Barros, Lillian; Calhelha, Ricardo C.; Heleno, Sandrina A.; Alves, Maria José; Carvalho, Ana Maria; Marouf, Abderrazak; Ferreira, Isabel C.F.R.The endemic Algerian myrtle, Myrtus nivelii Batt & Trab., was evaluated for its bioactive properties, such as antioxidant, anti-inflammatory, cytotoxic and antibacterial activities, and correlated with the individual phenolic compounds identified in its crude aqueous extract and subsequent organic fractions (ethyl acetate and butanol). Flavonols, such as myricetin, kaempferol and quercetin glycoside derivatives, were the major phenolic compounds found, along with the presence of ellagitannins. The ethyl acetate fraction contained the highest amount of phenolic compounds, followed by the butanol fraction and, finally, the crude aqueous extract. In general, all samples exhibited an excellent bioactivity, namely the ethyl acetate fraction that presented strong antioxidant activity, when compared to the standard trolox, strong cytotoxicity and antibacterial activity, especially against MRSA and MSSA. The present study revealed that the ethyl acetate fraction of M. nivelii could be used as a source of bioactive compounds in the food and pharmaceutical industries.
- Fingerprint of phenolic compounds in Osyris quadripartite Salzm. ex Decne. from AlgeriaPublication . The present study focused on the valorization of the leaves of Osyris quadripartite Salzm. ex Decne. with recognized folk medicinal uses in some Africa countries, namely Algeria [1, 2]. Therefore, the aim was to perform a phytochemical characterization, regarding phenolic composition, of O. quadripartita aqueous extracts and two different organic fractions (n-butanol and ethyl acetate), by using high-performance liquid chromatography coupled to a diode array detection and electrospray ionisation mass spectrometry (HPLC-DAD–ESI/MS). The separation was achieved with a Waters Spherisorb S3 ODS-2 C18 (4.6 mm × 150 mm, 3 μm), using a gradient elution, with 0.1% formic acid in water and acetonitrile. Twenty-eight individual phenolic compounds were identified: fifteen flavan-3-ols, six flavones, four flavonols, two phenolic acids and one flavanone derivative. The ethyl acetate fraction presented the highest concentration in phenolic compounds, being (+)-catechin and procyanidin dimer B1 (EC-4,8-C) the most abundant compounds (110.5 ± 0.3 and 100.5 ± 0.3 mg/g extract, respectively). The aqueous extract and the n-butanol fraction presented similar contents of phenolic compounds, being quercetin-3-O-rutinoside the main molecule present (33.8 ± 0.1 and 17.70 ± 0.02 mg/g extract, respectively). Moreover, the phenolic profiles of all fractions and extracts presented similarities in their qualitative composition, although some differences were observed, especially in the ethyl acetate fraction, where no flavone derivatives were present. This study highlights the potential of O. quadripartita fractions, rich in phenolic compounds, to be used in food, pharmaceutical and cosmetic fields.
- HPLC-DAD-ESI-MS/MS screening of phytochemical compounds and the bioactive properties of different plant parts of: Zizyphus lotus (L.) Desf.Publication . Rached, Wahiba; Barros, Lillian; Ziani, Borhane E.C.; Bennaceur, Malika; Calhelha, Ricardo C.; Heleno, Sandrina A.; Alves, Maria José; Marouf, Abderrazak; Ferreira, Isabel C.F.R.Zizyphus lotus L. (Desf.) has been widely used as a homemade ingredient to treat numerous diseases in Algerian folk medicine. In this context, decoction, infusion and hydroethanolic extracts were prepared from the branches, leaves, roots, and stem barks. The extracts were characterized regarding their phytochemical composition by HPLC-DAD-ESI/MSn and were assessed for in vitro bioactivity (cytotoxicity, anti-inflammatory activity, and antibacterial activity). A total of 29 compounds (flavonoid derivatives and secoiridoids) were identified in Z. lotus, 15 being detected in leaves, 10 in branches, 5 in roots and 7 in stem barks. The highest contents of these compounds, especially secoiridoids, were detected in branches followed by leaves, oleuropein being the main compound detected in leaves and oleoside in branches. The aqueous preparations of the leaves and root barks revealed the highest cytotoxicity and anti-inflammatory potential. On the other hand, as for the antibacterial activity, the hydroethanolic extract of the branches showed the highest potential, especially against the MSSA strain. Hence, the chemical diversity found in Z. lotus makes it a source of bioactive ingredients that can be applied in the formulations of different ingredients in the food and pharmaceutical industries.
- Phytochemical analysis and assessment of antioxidant, antimicrobial, anti-inflammatory and cytotoxic properties of Tetraclinis articulata (Vahl) Masters leavesPublication . The aim of this study was to evaluate the antioxidant, antibacterial, anti-inflammatory and cytotoxic properties of the aqueous crude extract and subsequent organic fractions of the Algerian medicinal plant T. articulata. Moreover, polyphenols were also fully profiled by liquid chromatography with diode-array detection coupled to electrospray ionization in tandem with mass spectrometry (LC–DAD–ESI–MSn). The analysis of the obtained results show that B-type (epi)catechin dimer and catechin were the most abundant phenolic compounds present among the nine different flavonoids identified such as: ((epi)catechin, myricetin, quercetin and kaempferol glycoside derivatives). The concentration of phenolic compounds in the ethyl acetate fraction (93.1 mg/g extract) was higher than the one of the aqueous extract (21.2 mg/g extract) and butanol fraction (43.87 mg/g extract). Furthermore, the ethyl acetate fraction revealed the strongest bioactive properties, revealing the lowest EC50/GI50 values: anti-inflammatory (EC50 129.67 μg/mL), cytotoxicity against human tumor cell lines (GI50, 59–189 μg/mL), antibacterial (MIC, 0.625–10 mg/mL); and especially the antioxidant capacity (EC50 values lower than 12.7 μg/mL), even higher than the standard (Trolox). The results showed a great bioactive potential for this species, with a significant contribution of flavonoid compounds, which makes it an interesting matrix in the development of novel pharmaceutical formulations.

An example is Osyris quadripartita, also known as Osyris lanceolata, which is known for its traditional therapeutic properties in some African countries. The aim of the present study was to evaluate the antioxidant (free radical scavenging activity, reducing power and lipid peroxidation inhibition), antiinflammatory (inhibition of NO production in lipopolysaccharide-stimulated RAW 264.7 macrophages) and cytotoxic (in a panel of human tumor cell lines and in non-tumor porcine liver primary cells) properties of O. quadripartita, providing a phytochemical characterization of its aqueous extracts and different organic fractions, by using high-performance liquid chromatography coupled to diode array detection and electrospray ionisation mass spectrometry (HPLC-DAD-ESI/MS). Twenty-eight individual phenolic compounds were identified: fifteen flavan-3-ols, six flavones, four flavonols, two phenolic acids and one flavanone derivative. The most abundant compounds in the ethyl acetate fraction were (+)-catechin and procyanidin dimer B1 (EC-4,8-C). Quercetin-3-O-rutinoside was the major compound in both the n-butanol fraction and crude aqueous extract. In most of the samples and assays the antioxidant activity was higher than the one revealed by the positive control gallic acid, with the highest antioxidant activity observed in the ethyl acetate fraction. The same fraction also showed the highest inhibition of NO production and the highest cytotoxicity against MCF-7 and NCI-H460 cell lines. This study highlights the potential of O. quadripartita fractions rich in phenolic compounds to be used in pharmaceutical and cosmetic fields.
