- ERBB2 in Cat Mammary Neoplasias Disclosed a Positive Correlation between RNA and Protein Low Expression Levels: A Model for erbB-2 Negative Human Breast CancerPublication . Santos, Sara; Batista, Claudia S.; Abreu, Rui M.V.; Bastos, Estela; Amorim, Irina; Gut, Ivo G.; Gartner, Fátima; Chaves, RaquelHuman ERBB2 is a proto-oncogene that codes for the erbB-2 epithelial growth factor receptor. In human breast cancer (HBC), erbB-2 protein overexpression has been repeatedly correlated with poor prognosis. In more recent works, underexpression of this gene has been described in HBC. Moreover, it is also recognised that oncogenes that are commonly amplified or deleted encompass point mutations, and some of these are associated with HBC. In cat mammary lesions (CMLs), the overexpression of ERBB2 (27%–59.6%) has also been described, mostly at the protein level and although cat mammary neoplasias are considered to be a natural model of HBC, molecular information is still scarce. In the present work, a cat ERBB2 fragment, comprising exons 10 to 15 (ERBB2_10–15) was achieved for the first time. Allelic variants and genomic haplotype analyses were also performed, and differences between normal and CML populations were observed. Three amino acid changes, corresponding to 3 non-synonymous genomic sequence variants that were only detected in CMLs, were proposed to damage the 3D structure of the protein. We analysed the cat ERBB2 gene at the DNA (copy number determination), mRNA (expression levels assessment) and protein levels (in extra- and intra protein domains) in CML samples and correlated the last two evaluations with clinicopathological features. We found a positive correlation between the expression levels of the ERBB2 RNA and erbB-2 protein, corresponding to the intracellular region. Additionally, we detected a positive correlation between higher mRNA expression and better clinical outcome. Our results suggest that the ERBB2 gene is post-transcriptionally regulated and that proteins with truncations and single point mutations are present in cat mammary neoplastic lesions. We would like to emphasise that the recurrent occurrence of low erbB-2 expression levels in cat mammary tumours, suggests the cat mammary neoplasias as a valuable model for erbB-2 negative HBC.
Introduction: A patient carrying ostomy is a person who has undergone surgery for diversion of intestinal and/or urine transit from normal path, due to a certain situation, whether disease, trauma or other. In this context, the nurse plays a very active role in trying to help and care for these patients that undergo drastic changes in relation to the representation of the body, in the practices, in the experiences, in family relationships, in sexual relationships and social relationships that affect work and leisure, reflecting on its autonomy (Borges et al. 2007). An adequate physical and psychological preparation is essential so that this patient recover its usual life, since it is a surgical procedure in which there are different feelings and different reactions like disbelief, denial, and shock (Fernandes, 2004). Objectives: To know the characteristics of users carrying ostomy disposal, to determine the degree of satisfaction with primary health care provided and to identify the teachings realized. Methods: An exploratory, cross-sectional and descriptive study was carried aout that involved by 43 individuals with elimination ostomies enrolled in Health Centers I and II from Bragança, Carrazeda de Ansiães and Miranda do Douro. For data collection, which took place from December 2012 to January 2013, it was used a questionnaire based on the literature review. The users were approached in their homes or at the consultation in their respective Health Centers. Results: Of the 43 users who participated in the study, the majority was male (60.5%), married or living with a partner (65.1%); was resident in rural areas (74.4%); lived with a spouse (62.8%); had a basic education (58.1%); and, was retired (86%). These patients were aged between 50 and 90 years old. Most patients were satisfied with the teachings acquired in what concerns to the ostomy hygiene and nutrition. Moreover, it was found that learning focused primarily on themes like the Ostomy Care, Food, Material to be used and suitable locations to purchase it. Discussion and Conclusion: The Health Education becomes increasingly important in nursing. The patient’s preparation, in physical and psychological aspects, is essential to recover his normal life.
- Improving Motor Competence of Children: The Super Quinas Intervention Program in Portuguese Primary SchoolsPublication . Rodrigues, Luis P.; Cordovil, Rita; Costa, Júlio A.; Seabra, André; Guilherme, José; Vale, Susana; Luz, Carlos; Flôres, Fábio; Lagoa, Maria João; Almeida, Gabriela; Lopes, Vitor P.; Mercê, Cristiana; Esteves, Pedro Tiago; Santos, Sara; Correia, Vanda; Serrano, João; Mendes, Rui; Matos, Rui; Loureiro, Vânia; Neto, CarlosThe objective of this study was to describe the effects of an extra hour of a structured motor program on the motor competence (MC) of children 6–10 years old. The need for movement interventions to enhance MC among school-aged children has gained vital importance in the last years, given the negative secular trends reported. Hence, the Portuguese Football Federation organized an intervention program on MC to be implemented on the extracurricular time of the Portuguese primary schools: the Super Quinas program. Thirty-nine schools from all of Portugal were assigned to intervention and control condition, with a total of 1034 children (6–10 y old) completing all the program (77.7%). The Super Quinas intervention comprised of 1 hour of activity per week, led by a physical educator teacher during extracurricular activities for 12 wees. MC was assessed using the Motor Competence Assessment (MCA) before and at the end of the program (January and April 2023). Normative results of the MCA were used to compare changes between pre and posttest according to experimental or control condition. Results showed a general improvement for all subscales (Locomotor, Manipulative, Stability) and total MCA. More importantly, the experimental group showed significant and positive differences, when compared with the control group, in the Stability Manipulative and total MCA results after controlling for gender, age, and baseline effect. Conclusions: The Super Quinas intervention program proved that adding 1 hour of structured movement program to the regular primary school schedule can lead to greater development of MC in school-age children.
