- Characterization of sour (Prunus cerasus L.) and sweet cherry (Prunus avium L.) varieties with five isozyme systemsPublication . Morales Corts, Remedios; Rodrigues, Luciano; Ortiz, Jesús María; Pérez Sánches, RodrigoExtracts from young leaves of nine sweet cherry (Prunus avium L.) and eight sour cherry (Prunus cerasus L.) varieties, located in the germplasm collection of the 'Direção Regional de Agricultura da Beira Interior' (Fundão, Portugal), were analysed for five isozyme systems in order to characterise these varieties and detect problems of synonymies and homonymies that frequently present. The sweet and sour cherry varieties analyzed showed low isoenzymatic polymorphism, being PGM and PGI the systems with the highest discrimination power. These systems presented seven and five different zymogrames, respectively. IDH showed four patterns. SKDH and 6-PGD grouped the varieties only into two patterns. The evident and discriminant restrictions of this type of analysis had got results that have only been a complement for agronomical and morphological characterization. Nove extratos de folhas com desenvolvimento encompleto foram obtidos de 9 variedades de cerejeira (Prunus avium L.) e 8 de ginjeira (Prunus cerasus L.), localizados no campo de germoplasma da "Direcção Regional de Agricultura da Beira Interior" (Fundão, Portugal). Esses extratos foram analisados para 5 sistemas isoenzimáticos a fim de caracterizar essas variedades e simultaneamente detectar sinonímias e homonímias, freqüentes neste tipo de estudos. As variedades de cerejeira e ginjeira analisadas mostraram baixo polimorfismo isoenzimático, sendo que os sistemas PGM e PGI mostraram maior poder de discriminação. Estes sistemas apresentaram 7 e 5 zimogramas diferentes, respectivamente. IDH mostrou 4 padrões. SKDH e 6–PGD agruparam as variedades em 2 padrões. Os evidentes limites discriminatórios desse tipo de análise serviram somente de complemento para a caracterização agronômica e morfológica.
- Expression analysis by RT-PCR of GIP gene from Phytophthora cinnamomiPublication . Belo, Hélio; Martins, Fátima; Jorge, Lurdes; Sousa, Maria João; Rodrigues, Luciano; Choupina, AltinoSpecies of the genus Phytophthora secrete glucanase inhibitor proteins (GIPs) to inhibit the activity of enzymes involved in plant defense responses, including during plant infection process of Castanea sativa Mill by Phytophthora cinnamomi. GIPs show structural homology to the chymotrypsin class of serine proteases (SP) but lack proteolytic activity due to the absence of an intact catalytic triad and, thus, belong to a broader class of proteins called serine protease homologs (SPH) nonfunctional because one or more residues of the essential catalytic triad is absent (His-Asp-Ser). GIPs show high homology to the S1A subfamily of SP, however questions remain about the expression patterns and potential roles of different GIPs during pathogenesis and their possible interaction with host EGases in the plant apoplast. ORF of GIP gene from P. cinnamomi encodes a 269 aa protein. In order to understand its function, we proceeded to the heterologous expression in Pichia pastoris. The expression was studied during growth in different carbon sources and a time course of glucanase inhibitor protein production by RT-PCR was also performed. The major expression levels occurred at the medium with glucose as carbon source.

- Morphological characterization of sweet and sour cherry cultivars in a germplasm bank at PortugalPublication . Rodrigues, Luciano; Remedios Morales, Maria; Fernandes, Artur João Bártolo; Ortiz, Jesús MaríaNine sweet cherry and eight sour cherry varieties located in a germplasm bank at Fundauo, Portugal, were studied from the viewpoint of characterization. Most of them were autochthonous cultivars that have a high risk of extinction since at the present they are markedly minor varieties. Morphological characteristics were evaluated in different organs: crown and trunk of the trees, leaves, flowers and fruits, over a three consecutive years period. Statistical analyses were carried out in order to detect similarities between cultivars as well as the existence of synonymies. Qualitative characteristics of the fruits were scored in order to carry out the multivariate analysis. A dendrogram of the evaluated characters shows the marked differentiation between sour and sweet cherries and suggests the existing synonymies. Conservation of the autochthonous cultivars in the future is highly recommended.
